CJC-1295 No DAC vs DAC: Pulsatile vs Sustained GH Release Research

Both versions of CJC-1295 activate the same GHRH receptor, but a single structural difference — the Drug Affinity Complex — produces two very different growth hormone release patterns in research.

CJC-1295 is a modified 29-amino-acid analogue of growth hormone-releasing hormone (GHRH), available in two research formats that share the same receptor target but produce mechanistically distinct release patterns: with, and without, a Drug Affinity Complex (DAC).

The DAC Difference

The Drug Affinity Complex is a chemical modification that binds serum albumin, dramatically extending the peptide's circulating half-life.

  • ·CJC-1295 (No DAC) — half-life of approximately 30 minutes, preserving a short, pulsatile GH release pattern that mimics the body's natural secretion rhythm
  • ·CJC-1295 (DAC) — half-life of approximately 6–8 days via albumin binding, producing sustained, elevated GH and IGF-1 levels across the entire dosing interval rather than discrete pulses

Both act on the same GHRH receptor at the anterior pituitary; the DAC modification changes nothing about receptor engagement, only how long the peptide remains available to trigger it.

Researchers studying the natural regulatory feedback of the GH axis — including negative feedback from somatostatin — often prefer the No DAC pulsatile pattern, since sustained non-pulsatile elevation can blunt some of the physiological feedback dynamics under study. Researchers more interested in steady-state IGF-1 exposure and dosing convenience often select the DAC variant.

Stacking with Ipamorelin

Both variants are studied in combination with Ipamorelin, a ghrelin (GHSR) receptor agonist that acts through a separate pathway. Reconstitution and Storage
