PT-141 (Bremelanotide): The Research Peptide for Sexual Function
PT-141 works through the central nervous system rather than the vascular route of most libido compounds. Here's the melanocortin science, the research data, and how it's handled.
PT-141, also known as Bremelanotide, is one of the more unusual peptides in sexual-health research. Unlike compounds that act on blood flow, PT-141 works in the brain through the melanocortin system — making it relevant to research on desire rather than mechanics alone.
What Is PT-141?
PT-141 is a synthetic cyclic heptapeptide derived from Melanotan II. Researchers studying Melanotan II's tanning effects noticed it also produced arousal responses. PT-141 was developed by isolating that pathway, removing the pigmentation-driving activity while retaining the effect on sexual function.
How Does PT-141 Work?
PT-141 is a melanocortin receptor agonist, acting primarily on the MC4 receptor in the central nervous system. This distinguishes it from PDE5-based approaches:
- ·Central mechanism — activates melanocortin pathways in the hypothalamus rather than acting on peripheral blood vessels
- ·Desire-oriented — research targets libido and arousal signalling, not just physical response
- ·Route-independent of vascular function — because it does not rely on the nitric oxide/vascular pathway, it has been studied in models where vascular-acting compounds are less effective
Because the mechanism is central, PT-141 has been researched in both male and female models of sexual dysfunction.
What Does the Research Show?
Bremelanotide reached clinical development for hypoactive sexual desire disorder in premenopausal women and was approved by the FDA in that indication under a separate brand. Trials reported improvements in desire and reductions in distress versus placebo. Male-model research has examined arousal and erectile response, particularly in cases unresponsive to vascular-acting agents.
PT-141 vs Melanotan II
The two are chemically related but researched for different endpoints:
- ·PT-141 — sexual function via MC4 receptor; minimal pigmentation activity
- ·Melanotan II — melanogenesis (tanning) via MC1 receptor, with libido as a secondary effect
Researchers interested primarily in arousal endpoints typically choose PT-141 for its cleaner selectivity. See our Melanotan II listing for comparison.
Reconstitution and Handling
PT-141 is supplied as a lyophilized powder and reconstituted with bacteriostatic water before use. Store the powder cold and dark; refrigerate after reconstitution and protect from light. Our reconstitution guide covers the full procedure.
