TB-500 is a synthetic peptide fragment corresponding to the active region of thymosin beta-4 (Tβ4), a naturally occurring 43-amino-acid protein involved in tissue repair and regeneration. Its primary mechanism is G-actin sequestration, regulating the actin polymerisation critical for cell migration, division, and wound closure. TB-500 promotes angiogenesis through VEGF upregulation, inhibits NF-κB inflammatory signalling, and mobilises stem cells to injury sites. Its low molecular weight enables systemic distribution, meaning injection site is less critical than with locally-acting peptides.
