SS-31 (also known as elamipretide or Bendavia) is a small, cell-permeable tetrapeptide engineered specifically to concentrate inside mitochondria — a design that sets it apart from most other research peptides, which act at cell-surface receptors rather than penetrating directly into an organelle.

Design and Targeting Mechanism

SS-31 was designed by alternating cationic and aromatic amino acid residues, giving it two key properties:

  • ·Positive charge — draws it toward the highly electronegative inner mitochondrial membrane
  • ·Aromatic-cationic motif — allows it to concentrate in mitochondria at levels roughly 1,000-fold higher than in the surrounding cytosol, independent of the mitochondrial membrane potential

This targeting mechanism means SS-31 accumulates precisely where oxidative damage originates, rather than acting diffusely throughout the cell.

What SS-31 Does Inside the Mitochondria

The peptide's primary documented mechanism is binding to cardiolipin, a phospholipid unique to the inner mitochondrial membrane that is essential for organizing the electron transport chain complexes.

  • ·Cardiolipin stabilization — prevents cardiolipin peroxidation, which otherwise destabilizes the cristae structure and impairs ATP synthesis
  • ·Electron transport chain efficiency — preclinical research shows improved coupling of oxidative phosphorylation, meaning more ATP is generated per unit of oxygen consumed
