5-Amino-1MQ is not a peptide — it is a small molecule — but it has become a fixture in the metabolic and longevity research space because of an unusual mechanism: it raises cellular NAD+ from a completely different angle than the popular NAD+ precursors.

What Is 5-Amino-1MQ?

5-Amino-1MQ (5-amino-1-methylquinolinium) is a selective inhibitor of the enzyme NNMT (nicotinamide N-methyltransferase). NNMT is overexpressed in adipose tissue and in some aging and metabolic-disease states. By inhibiting it, 5-Amino-1MQ has been studied for its effects on fat cell metabolism and NAD+ availability.

How Does 5-Amino-1MQ Work?

NNMT consumes methyl groups and NAD+-related substrate to methylate nicotinamide. When it is overactive, two things happen: cellular energy metabolism slows in fat tissue, and NAD+ salvage is impaired. Inhibiting NNMT has been researched for the reverse effects:

  • ·Raises cellular NAD+ — frees up nicotinamide for the NAD+ salvage pathway rather than shunting it to methylation
  • ·Increases adipocyte energy expenditure — in preclinical models, blocking NNMT reduced fat cell size and increased metabolic rate
  • ·Distinct from NAD+ precursors — precursors like NMN and NR supply raw material; 5-Amino-1MQ instead removes a drain on the system

What Does the Research Show?

Preclinical studies reported that NNMT inhibition reduced adipose tissue mass and improved metabolic parameters in diet-induced obesity models, without reducing food intake — pointing to an energy-expenditure mechanism rather than appetite suppression. This contrasts with GLP-1 agonists like Semaglutide, which work primarily by reducing intake. Much of the 5-Amino-1MQ data is still preclinical, and human research is early.
